Exploring Microscopy Techniques in Bio/Life Sciences: Challenges, Strategies, and Alternatives

<p xmlns:xsi="http://www.w3.org/2001/XMLSchema-instance" class="first" dir="auto" id="d18261e44">Scanning and transmission electron microscopy (SEM and TEM) are crucial in bio/life sciences, providing invaluable insights into biological sample ultrastructure. Alongside SEM and TEM, various microscopy techniques are utilized for studying specimens across scales. This review examines challenges inherent in SEM, TEM, and other methods, alongside preparation strategies to mitigate them. Challenges include sample preparation, resolution limitations, artifact formation, beam damage, and contrast enhancement. Preparation methods like fixation, dehydration, and cryo-fixation address these challenges, preserving sample integrity and enhancing contrast. Additionally, alternative microscopy techniques such as multiphoton microscopy, confocal microscopy, and fluorescence microscopy are discussed, each with unique advantages and challenges in sample preparation, resolution, and cost. Proper technique selection and optimization, coupled with meticulous sample preparation, are crucial for acquiring reliable data and advancing biological understanding.
