Abstract 2476 The Molecular basis for the Mechanism of IFI16β oligomerization and DNA binding

AIM2 and IFI16 are inflammasome sensor proteins that trigger inflammation in response to DNA. These proteins contain a DNA binding domain (HIN) and an oligomerization Pyrin Domain (PYD) for signal activation. The overactivation and dysregulation of inflammasomes can lead to autoimmune and autoinflammatory diseases, and therefore, they must be tightly regulated. A newly classified isoform of human IFI16, known as IFI16β, regulates the inflammatory response of AIM2. IFI16β contains the two HIN domains of IFI16 but lacks the PYD oligomerization domain for inflammatory signaling.
