Characterization of intestinal dendritic cells and macrophages of active hexose correlated compound-fed stressed mice during Chlamydia muridarum genital infection 3526

Abstract Description Active hexose correlated compound (AHCC), an extract of mushrooms commonly used as a dietary supplement, boosts the immune system. The objective of this study was to characterize the profiles of dendritic cells (DCs), and macrophages (MΦ) isolated from intestine of stressed BALB/c mice fed with AHCC during Chlmaydia muridarum genital infection. Antigen-presenting cells, DCs and MΦ play critical roles in delivering the AHCC immunogenic particle to T cells, but their phenotypes and function in our mouse model are not known. The phenotypic profiling of DCs and MΦ subsets using flow cytometric analysis is underway. Two major populations, CD11c and CD11b, were analyzed for the expression of identifying surface markers. A two-fold increased expression of CD103 (7.16%) in AHCC-fed compared to phosphate-buffered saline(PBS)-fed mice was observed. Similarly, CD45R/B220 was 48.29% in AHCC-fed compared to (35.23%) in PBS-fed mice. In MΦ, CD11b+CD64a expressed was 53.69 in AHCC-fed cells compared to 29.34 % of CD11b+CD64a expression in PBS-fed mice. Expression of F4/80 in AHCC-fed mice was two-fold (13%) higher than in PBS-fed mice. Flow cytometric approach to identify the effect of AHCC in the expression of DC and MΦ subsets in mice during chlamydia genital infection will provide a useful tool to analyze the immune response of mice during chlamydia genital infection.
