Enhancing workflows for Cytokine Profiling and Protein Analysis using a consistent, convenient and high-throughput platform – PIXelated ULtrasound (PIXUL®) 4565

Abstract Description Cytokines are secreted by diverse cells as small proteins which regulate immunology, development and homeostasis. They are implicated in autoimmune and infectious diseases, chemotaxis, cell communication, and immune cell differentiation during inflammation. Hence, accurate detection and analyses of cytokine profiles are crucial. However, several challenges exist with traditional cytokine profiling like low sample content from patients, assay sensitivity, detection range, accuracy, throughput, costs and labor. Here, we address the above challenges by integrating cavitation-based PIXUL® to cytokine profiling and protein analyses. Results show that using limited quantity input, PIXUL® prepares 96 samples in 5 minutes only, with higher consistency and reliability compared to traditional methods requiring hours. It integrates with MSD’s single-/multi-plex phosphoprotein analyses, custom multiplex analyte-combination profiling, automated protein separation-immunodetection and more. PIXUL®’s compatibility with cells, tissues, other biological samples, and its economical labware enabling 2D and 3D cell cultures and their processing, together enable such wide range of applications. Cytokine profiling informs disease progression, severity, outcomes, signatures, and interventions like CAR T cell therapy. Our simple and accessible technology will enable academia, pharma, clinical trials, personalized medicine and companion diagnostics to analyze diagnostic and predictive biomarkers.
