Mouse IgG2c Ly6G [1A8]: A new chimeric recombinant antibody for improved neutrophil depletion In Vivo 9117

Abstract Description Neutrophils play a critical role in immune defense, wound healing, autoimmunity, and cancer. To study their function in vivo, antibody-mediated depletion of neutrophils by targeting the neutrophil-specific Ly6G marker is commonly used. The commonly used rat IgG2a hybridoma antibody clone 1A8 specifically targets Ly6G, inducing neutropenia for 2 days, necessitating repeated injections for long-term studies. To improve efficacy, reproducibility, and animal welfare, we developed a recombinant murinized version of the Ly6G [1A8] clone, extending its half-life by eliminating issues caused by anti-rat antibody production. Similarly, we created a chimeric mouse IgG2c anti-Ly6G [1A8] antibody variant which was validated for specificity and tested in vivo using BigRed/CatchupIVM-red mice. Results showed that mouse IgG2c anti-Ly6G [1A8] provides sustained neutrophil depletion for 72 hours reducing the number of required injections compared to the original hybridoma rat IgG2a. Additionally, downregulation of Ly6G on remaining neutrophils was observed, indicating new bone marrow-derived neutrophils were generated. Abcam’s new chimeric mouse IgG2c anti-Ly6G [1A8] antibody (ab320708) improves animal welfare and simplifies experimental protocols by reducing injection frequency.
