Enhanced B cell immune responses in humanized γδ TCR mice post antigens immunization 2643

Abstract Description The humanized gamma delta (γδ) T cell receptor (TCR) mouse model represents a pivotal advancement in immunological research, enabling more accurate study of human immune responses. This study evaluates the B cell-mediated immune responses in humanized γδ TCR mice and compared it to wild type (WT) mice after immunization with collagen and keratin immunizations on B cell immune responses in humanized γδ TCR mice and WT mice. Our findings reveal that these immunizations significantly enhance B cell activity in HuTCR mice, as evidenced by increased numbers of antigen-specific antibody-secreting cells (ASCs), elevated antibody titers, and higher percentages of plasma and memory B cells in systemic tissues compared to WT mice. Additionally, we observed pronounced elevated levels of both immunoregulatory and pro-inflammatory cytokines in the serum. These results suggest that γδ TCR signaling plays a critical role in modulating B cell responses, offering new insights into the interplay between γδ T cells and B cells in the context of immunization. This enhanced understanding of γδ TCR-mediated B cell activation could pave the way for novel immunotherapeutic strategies targeting autoimmune diseases and improving vaccine efficacy.
