hIL4/hIL4R mice as a asthma model induced by HDM enable to assess the efficacy and safety of therapies in vivo 3210

Abstract Description Background Asthma is a chronic inflammatory disease of the airways characterized by obstruction [1]. House dust mites (HDM) are commonly used to model asthma in experimental settings. Dupilumab has been approved for patients with uncontrolled moderate-to-severe asthma [2]. This study aimed to evaluate the ability of HDM to induce asthma in hIL4/hIL4R mice and to assess the efficacy of dupilumab in controlling this condition. Methods The asthma was induced by HDM. The 6-week-old mice were randomly divided into three groups on Day 1.The G1 group served as controls, while groups G2-G3 received 30 μg of HDM. Mice in the G3 group received the Dupilumab treatment. At endpoint, flow cytometry analyzed the proportion of eosinophils in alveolar lavage fluid. ELISA detected the serum IgE level. Histopathological analysis assessed inflammation and mucus formation in the bronchi. QPCR detected the levels of inflammation factors. Conclusion The HDM model demonstrated significant pathological differences compared to control group, including the increased proportions eosinophils in BALF, the level of IgE, the inflammatory and mucus in the lung. Treatment with dupilumab effectively improved the pathological scores, reduced circulating IgE level, and down-regulated the eosinophils and inflammtion factors in the lung organ. Overall, the hIL4/hIL4R mice could successfully induced by HDM as asthma model and dupilumab assessing the efficacy and safety of potential therapeutic interventions.
