6P Natural IgM and dyslipidemia enhance the efficacy of tumor necrosis factor and interleukin-2 encoding oncolytic adenovirus in solid tumors

The oncolytic adenovirus TILT-123 has demonstrated a favourable safety profile and durable responses in phase I trials for advanced solid tumours. To support clinical translation, identifying biomarkers that predict intravenous (i.v.) delivery is critical. Innate immune components, including natural IgM antibodies, which bind pathogens and modified low-density lipoprotein (LDL), affect i.v. adenoviral therapy by directing viral particles to liver Kupffer cells, reducing systemic delivery and efficacy.
