Photo‐Controlled Antibacterial Drug Release From DASA‐Coated Silica Particles for Improved Treatment of Wound Infections

ABSTRACT Infected chronic wounds typically require complex management and daily care, resulting in a substantial socioeconomic burden. Precise and controllable drug delivery systems can reduce the need for frequent medical interventions, lowering associated costs while maximising the effectiveness of chronic wound treatments. Conventional drug delivery systems can provide sustained release but lack the spatiotemporal control needed to limit side effects. In this work, we report the synthesis of donor–acceptor Stenhouse adduct‐functionalised, chlorhexidine‐loaded silica particles that enable light‐controlled release of antiseptic compounds. Kinetic studies demonstrated a light‐modulated release mechanism triggered by green light irradiation, independent of wound‐relevant environmental parameters, such as pH. Bacterial studies showed controlled antimicrobial activity against both Staphylococcus aureus and Escherichia coli , with linear reductions in bacterial load to 6% and 23%, respectively, following irradiation. Human dermal fibroblasts exhibited good viability in the presence of the particles, further supporting their suitability for controlled local treatment of infected wounds. These findings pave the way toward personalised treatment of chronic wound infections using light‐responsive silica particles to achieve on‐demand drug delivery.
