Biopolymer carriers of polydeoxyribonucleotides for correction of impaired healing of thermal burns

Objective. To analyze the mechanisms of regenerative and anti-inflammatory potential of polydeoxyribonucleotide-containing drugs using a thermal burn model in experimental animals. Material and methods. Preclinical trials of regenerative and anti-inflammatory potential of Nucleoform gel, containing 2% polydeoxyribonucleotide solution, were conducted using a thermal burn model in Wistar rats. The control group of animals received no treatment, while the comparison group received Levomekol ointment. Results. Dorsal skin burns over an area of approximately 300 mm2 are accompanied by high activity of proinflammatory cytokines (interleukin 1β and tumor necrosis factor-alpha) due to imbalance in lipid peroxidation system. In the control group, burn area decreased by 38% over 20 days, in the comparison group 0 by 51.2%, in the study group — by 63.4%. These changes were accompanied by lower inflammatory activity, especially under Nucleoform gel application. Similar trends were observed in lipid peroxidation system. Correlation analysis showed that thermal burn significantly enhanced relationship between all parameters, but this phenomenon gradually disappears with therapeutic intervention, particularly with DNA derivatives. Conclusion. Experimental results indicate significant therapeutic potential of polydeoxyribonucleotide-based drugs in human burns.
