Greenness, Whiteness, and Sustainability Profiling of an Ethanol-BasedRP-HPLC Method for Atorvastatin and Clopidogrel in PharmaceuticalTablets

Atorvastatin calcium and clopidogrel bisulfate were regularly prescribed together in a fixed dose combination for a wide range of cardiovascular therapies. A thorough literature review of existing methods suggested that quantifying this combination depends highly on acetonitrile, which is a hazardous solvent. Green analytical chemistry helps to minimise the use of solvents and replace conventional solvents with green solvents to explore the greener and safer analytical performance. In the current study, a greener method was developed with a Waters Xterra RP18 column and a mobile phase of 10mM potassium phosphate buffer in the ratio of 70:30 v/v. The flow rate was set to 1.2 mL/min, the run time was 6 minutes, and the wavelength was 230 nm. Both the drugs expressed good separation with a retention time of 2.152 minutes and 3.646 minutes. The linearity was within the limits with the regression values of 0.9998 and 0.9996. The precision stayed below 1% RSD, and the accuracy was also within the allowed limits. The tablets showed drug content close to the label. Some green tools were checked to see the environmental impact. They all showed that the method is greener because ethanol was used instead of acetonitrile. The method is simple and steady and can be used easily for routine work.
