Aminopyrifen and Other <scp>GPI</scp> ‐anchored Wall Transfer Protein Inhibitors

A glycosylphosphatidylinositol wall transfer protein1, Gwt1, is localized on the endoplasmic reticulum membrane and is involved in the early step of GPI biosynthesis. GPI anchors mannoproteins to form cell walls at the plasma membrane, which is essential for cell wall assembly in yeasts and molds. Medicinal chemistry research identified Gwt1 as a target molecule for antifungal and generated Gwt1 inhibitors such as BIQ and manogepix. On the other hand, aminopyrifen, with a unique 2-aminonicotinate, was discovered through drug discovery research in crop protection. Consequently, failure of GPI biosynthesis by those inhibitors attenuates the pathogenicity of clinical and agricultural pathogens through cell wall defects. Gwt1 inhibitor is a promising tool for controlling harmful pathogens in clinical and agricultural situations.
