The Minimum TCR-pMHC Threshold for T Cell Activation Is N=3: First-Principles Derivation from the Cooperative Percolation Constant ln(3)

Abstract: T cell activation historically requires a minimum of approximately 3 sustained TCR-pMHC engagements. While single ($N=1$) or paired ($N=2$) contacts yield low activation probabilities (<20%), a sharp transition occurs at $N=3$ (>68%). This 30-year immunological mystery lacks a unified theoretical explanation. We derive $N_{min} = k = 3 \approx e^{\ln(3)}$ from first principles without free parameters. Based on the Cooperative Percolation Constant $\ln(3)$ (arXiv:2603.15521), we demonstrate that TCR micro-clusters migrating along quasi-linear paths (1D process) require bidirectional signal coordination to trigger stable downstream signaling. In a 1D Poisson process, the minimum cluster size for such coordination is $k = e^{\ln(3)} \approx 3$. Validation: Our model is validated against three landmark experimental datasets: (1) Irvine et al. (2002, Nature), (2) Huang et al. (2013, Immunity) using DNA origami, and (3) Bhatt et al. (2019, eLife). We further propose an optimal CAR-T construct spacing $d^* \approx 9\text{ nm}$ based on $l_{CAR}/\ln(3)$.
