Effects of Bactrian camel exosome miR-144-5p on liver inflammation in obese mice

ABSTRACT To explore the effect of Bactrian camel exosome miR-144-5p on liver in obese mice forty male mice were randomly divided into a control group (n=10) and a obesity test group (fed high-fat chow, n=30). The obesity group was further divided into model group, miR-144-5p agomir and miR-144-5p antagomir. Mice in each group were injected with saline, miR-144-5p agomir, and miR-144-5p antagomir by tail vein once every 3 days for 7 times, respectively. At the end of the experiment, serum and liver tissues of mice in each group were collected. The levels of tumor necrosis factor-α (TNF-α) and arginase-1 (Arg-1) in serum were detected by ELISA. The expression levels of CPT1-α、FASN、TNF-α and Arg-l in liver tissue were detected by QRT-PCR. The expression levels of TNF-α and Arg-l protein in liver tissue were detected by Western Blot. The results showed that compared with the model group, the miR-144-5p agomir group could inhibit the expression of TNF-α and promote the expression of Arg-l, while the miR-144-5p antagomir group promoted the expression of TNF-α and inhibited the expression of Arg-l. Therefore, miR-144-5p may reduce the inflammatory response in the liver of obese mice by inhibiting inflammatory factors and promoting the expression of anti-inflammatory factors.
