3,4-Dihydroxyamphetamine (HHA) and 3,4-dihydroxymethamphetamine (HHMA) are the principal catecholic metabolites of MDMA ("ecstasy") and are required as certified reference standards (CRMs) for GMP-grade bioanalytical methods in clinical pharmacology and forensic toxicology. Their structural combination of an ortho-catechol moiety with a proximal unhindered aliphatic amine drives an irreversible oxidative cascade — autoxidation, intramolecular aza-Michael cyclisation, and aromatisation to melanin-type polymers — that renders both compounds acutely unstable under ambient conditions. This note describes the synthetic strategy (dibenzyl ether protection / catalytic hydrogenolysis under inert atmosphere), the purification approach (RP-8 MPLC with 1% aqueous HCl / isopropanol), and the critical handling conditions (pH ≤1, inert atmosphere, −20°C) required to obtain and maintain these metabolites in analytically useful form. A mechanistic rationale for each precaution is provided. The relevance to ongoing GMP clinical investigations of MDMA-assisted therapy and to ICH M10-compliant bioanalytical method validation is discussed.
