Evaluation of Biochemical, Physiometric, Anthropometric and in-silico analysis of Cyp11b2 gene in Diabetics, Cardiovascular diseases in north Indian population: A case-control study

Aim: The study was conducted an evaluation of Biochemical, Physiometric, Anthropometric and in-silico analysis of Cyp11b2 gene in Diabetes, Cardiovascular diseases in north Indian population. This is the first extensive molecular analysis in northern India. Methods: It was determined in 50 diabetes, 50 cardiovascular, 50 diabetes with cardiovascular and 50 normal individuals. The relationship of anthropometric, lipid profile and blood pressure were determined by student t-test and physiometric analysis was determined by chi square test. It concluded that Hp2-2 may be a marker of baseline characteristics and serum levels in individuals having diabetics, cardiovascular disease and cardiovascular with diabetics. Results: The Anthropometric, Physiometric and Biochemical parameters was statistically significant in diabetes as compared to normal participants and physiometric parameters was statistically significant in diseases as compared to normal participants i.e. p<0.05. Lipid levels was not statistically significant in diabetes as compared to normal participant i.e. p>0.05 but in other variables it was statistically significant i.e. p<0.05. A total of 15 SNP’s of CYP11b2 were extracted randomly from NCBI database with a SNP’s MAF value ≤0.05. The physical interaction of CYP11b2 with other genes was 77.74%. The mutations effect of SNP’s predicting by using FuncPred, SIFT, PolymiRTS and Regulomedb database. A 6 SNPs variants were obtained to be affecting transcription factor binding site by FuncPred tool and one SNP at 3’UTR i.e. rs28491316 was found to affect miRNA binding site. one SNP’s rs141179680 had very likely to affect the binding for TF binding + matched TF motif + matched DNase footprint + DNase peak (2b) and 3 SNPs had regulomedb score of ‘1f’ elucidation for eQTL+ TF binding DNase peak and thus expected to have regulatory functions. Conclusion: Body mass Index, Waist circumference and waist hip ratio is a risk factor of obesity, diabetes and cardiovascular disease in our study. Atherogenic index plasma is not showing a correlation only in diabetic mellitus individuals as compared to normal participant in our study. 5 SNP’s of Cyp11b2 were found deleterious or serious effect, 9 SNP’s showed tolerated effect with a score of 1, 0.35, 0.75, 0.78 and 0.34. If values vary from 0 to 1 is predicted to effect the function of protein. Some SNP’s were found mutation in the region of promoter that can alter the transcription of gene by various computational tools. This will become more relevant in the next-generation sequencing data analysis to study any type of disease
