Unveiling the Cosmetic Potential of Umbilical Cord-Derived Mesenchymal Stem Cell Secretome: A Proteomic and Functional Characterization for Skin Regeneration

Introduction: Umbilical cord-derived mesenchymal stem cells (UC-MSCs) have emerged as a promising source of regenerative bioactive compounds, positioning them as valuable candidates for use in dermatological and cosmetic formulations. Materials and Methods: In this study, the secretome (conditioned media) of UC-MSCs (MSCs-CM) was comprehensively characterized for its suitability as an active cosmetic ingredient for skin and hair. UC-MSCs were validated for mesenchymal identity by assessing morphology, surface marker expression, and trilineage differentiation. Results: The MSCs-CM demonstrated significant bioactivity, including inhibition of key enzymes associated with oxidative stress, collagen degradation, and pigmentation. Additionally, MSCs-CM promoted wound healing and preserved cellular viability across varying doses. Proteomic analysis identified 163 high-confidence proteins in the secretome, with functional enrichment showing predominant roles in extracellular matrix organization, immune modulation, cell signaling, and regeneration. Discussion: Gene ontology analysis and network-based mapping highlighted the extensive involvement of proteins in extracellular vesicle biology, cytoskeletal dynamics, and antioxidant responses. These functional attributes support the cosmetic relevance of the UC-MSC-derived secretome, particularly for addressing aging, pigmentation, and tissue repair. Conclusion: Overall, the regenerative, antioxidant, and dermo-cosmetic properties identified in the UC-MSC secretome underscore its potential as an innovative active ingredient for advanced cosmeceutical formulations targeting skin and hair rejuvenation.
